At some point, we will have exhausted Earth's resources and mankind will need to expand the reaches of human civilization to survive. To ensure we're ready for that, we need to learn more about the effects of long duration flight on the human body and psyche. We also need to develop self-sustaining life support systems that will reduce, and eventually eliminate, the need for re-supply from Earth, allowing us to complete spaceflights of a thousand days and more.
